In 1954, while Margot Eskens was working as a dental assistant, she triumphed in a talent contest held by Polydor Records with the track "Moulin Rouge." After that, she signed a deal with Polydor, and in 1955, she achieved her debut success in Germany with the song "Ich mochte heut ausgehn."She had two number one singles in 1956 and 1957: "Tiritomba," which sold more than 800,000 copies, and "Cindy oh Cindy," which was her greatest hit and ran for 25 weeks in the German Singles Top 10.Following that, there were a few duets performed with Silvio Francesco [de], who is singer Caterina Valente's brother."Himmelblaue Serenade" is a single which she recorded with Silvio Francesco. Up to the year 1961, Margot Eskens worked closely with the producer and lyricist Kurt Feltz. In 1962, she competed in the German Schlager-Festspiele with "A heart, you can't buy that" and achieved the third spot, behind only Conny Froboess and Siw Malmkvist. She competed in the Eurovision Song Competition as a representative of Germany, in 1966 with the song "The Hands of the Clock" and finished in the tenth spot. Although she had little chance against her erstwhile duet partner UDO JÜRGENS ("Merci Cherie"), who was singing for Austria. This was nevertheless a little triumph for Germany because the German entries since 1964 and 1965 were not awarded any points. Since 1956, she has amassed 40 million record sales. She started to make more conventional "Schlagermusik" towards the end of the 1980s, such as her album "Mein Traumland am Worthersee" in 1990.

Duets were performed by Margot Eskens with a number of well-known artists, such as René Carol ,  Udo Jürgens, Peter Alexander, Will Brandes, Silvio Francesco and Willy Hagara to name just a few.
